of Dissertation Presented to the Graduate School of the University of Florida in Partial Fulfillment of the Requirements for the Degree of Doctor of Philosophy DIFFUSION MAGNETIC RESONANCE IMAGING IN A RAT HIPPOCAMPAL SLICE MODEL OF ACUTE BRAIN INJURY By Timothy Michael Shepherd May 2003 Chair: Edward D. Wirth, III Major Department: Neuroscience This dissertation describes experiments using rat hippocampal slices as a model of nervous tissue to evaluate the changes in diffusion-weighted magnetic resonance images (MRI) observed after acute brain injury. A multislice perfusion chamber was designed to acquire diffusion-weighted MRI from multiple rat hippocampal slices simultaneously. These images had signal-to-noise ratios sufficient to analyze the multicomponent diffusion properties of water in rat hippocampal slices. Because the hippocampal slice water diffusion parameters were stable for at least 8 hours after procurement in this chamber, it enabled studies of the temporal evolution of diffusion changes in slices following perturbation with the calcium ionophore A23187. The mean fast diffusing water fraction (Ffast) progressively decreased for slices treated with 10 μM A23187 (-20.9 ± 6.3% at 8 hours). Slices treated with 50 μM A23187 had significantly reduced Ffast 80 minutes earlier than slices treated with 10 μM A23187 (P < 0.05). These data suggested diffusion-weighted images at high b-values and the diffusion parameter Ffast may be

MR microscopy of rat hippocampal slice cultures: a novel model for studying cellular processes and chronic perturbations to tissue microstructure.
Brain slices provide a useful nervous tissue model to investigate the relationships between magnetic resonance imaging (MRI) contrast mechanisms and tissue microstructure; yet, these acutely isolated tissues remain viable for only 10-12 h. To study slower biological processes, this work describes the first MRI microscopy characterization of organotypic rat hippocampal slice cultures that can be...
متن کاملClinical Significance of Diffusion Weighted Magnetic Resonance Imaging on Treatment Efficacy in MS Patients With Acute Attacks
Background: Despite the existence of various imaging methods, the accurate diagnosis of many neurodegenerative diseases is still controversial. The use of advanced imaging techniques, such as the diffusion weighted imaging can be help the early detection of MS and evaluation of the treatment efficacy in these patients. Materials and method: 24 MS patients with acute attack and thirty healthy s...
متن کاملStructural Findings in the Brain MRI of Patients with Acute Carbon Monoxide Poisoning
Background: Carbon monoxide (CO) poisoning may lead to hypoxic/anoxic injury and eventually ischemic encephalopathy. Magnetic resonance imaging (MRI) has a well-recognized role in assessment of the severity of brain damage caused by CO poisoning. In this study, we aimed to present and analyze the structural abnormalities in the brain MRI and especially in diffusion weighted MRI (DWI) images in ...
متن کاملDetection of Alzheimer\'s disease based on magnetic resonance imaging of the brain using support vector machine model
Background: Alzheimer's disease (AD) is the most common disorder of dementia, which has not been cured after its occurrence. AD progresses indiscernible, first destroy the structure of the brain and subsequently becomes clinically evident. Therefore, the timely and correct diagnosis of these structural changes in the brain is very important and it can prevent the disease or stop its progress. N...
متن کاملThe effect of ouabain on water diffusion in the rat hippocampal slice measured by high resolution NMR imaging.
High resolution NMR imaging of the isolated perfused rat hippocampal slice was used to quantitate ADC changes following ouabain-induced cell swelling. Hippocampal slices were studied in artificial cerebrospinal fluid and then in ouabain using a 600-MHz narrow bore spectrometer and a home-built perfusion chamber. The brain slices demonstrated biexponential diffusion behavior. After perfusion wit...
